Are American Airlines' new Boeing 777-300ERs going to be late into ...

We?re hearing that American Airlines is having the problems with its soon-to-arrive Boeing 777-300ER aircraft. In particular, we?re hearing that the problems are with the fancy first-class seats being built in Great Britain.

If so, that would threaten the aircraft?s planned entry into service in American?s schedule, on the Dallas/Fort Worth-Sao Paulo, Brazil, route beginning Dec. 13.

We talked Tuesday to American?s chief commercial officer Virasb Vahidi about American?s plan to launch some new international routes. While we had him on the phone, we also asked if American had solved the problem with the first-class seats, whether American would be able to put the airplane into service as planned.

His reply: ?Well, stay tuned. We?re probably pretty close to making an announcement on the exact entry into service of that aircraft very soon.?

We pointed out that American had already announced that 777-300ER would begin flying the DFW-Sao Paulo route on Dec. 13.

His reply: ?That is our current stance or current position and that?s what?s in the GDS?s or the CRS?s and that?s what the plan is as?of right now. That?s correct.? (GDS=Global distribution systems, CRS=computerized reservations systems.)

We asked whether that start date is in jeopardy.

His reply: ?That is the stake in the ground and plan of record as of right now. If that changes, we?ll let everyone know, including our customers.?

At that point, a public relations person interrupted to say that Virasb had other interviews he needed to do, and that ended the questioning.

When it goes into service, the 777-300ER will be the biggest aircraft that American operates. It has, if we counted right, 310 seats, compared to the 777-200?s 247 seats. The 777-200 currently flies the D/FW-Sao Paulo route, as well as a number of other important international routes.

American has 14 announced orders for the 777-300ER, with two to arrive this year, eight coming in 2013 and four more in 2014.

On Nov. 30, 2011, American announced the details of the 777-300ER?s interior, and made much of the first-class seats:

?Considered one of the most comfortable and popular First Class seats in the industry, the 777-300 will feature an updated and enhanced version of the customer-favorite Flagship Suite seat that transforms into a fully lie flat six-foot, eight-inch bed with drop-down armrests that allow for easier access and more sleeping space.

?Each suite provides aisle access and allows customers to individually adjust any component of the seat for ultimate comfort. All suites will feature privacy dividers, two universal AC power outlets, one USB outlet, two large tray tables, a swivel seat and a host of other comforts, including Bose QuietComfort 15 Acoustic Noise Cancelling headsets, designed to enhance the travel experience.

?A 17-inch touchscreen monitor will offer an array of entertainment and a touchscreen video handset will serve as a remote that allows customers to easily select programming without touching the monitor and watch additional video programs, such as the moving map, while viewing the 17-inch monitor. The First Class cabin will use updated interior trim and finishes to achieve a comfortable, modern feel throughout the cabin.?

At that time, American said London would be ?a first market? for the airplane. But on May 19, it announced that the D/FW-Sao Paulo route would get the first 777-300ER on Dec. 13, followed by D/FW-London on Jan. 31, New York-London in February, a second D/FW-London flight in March and New York-Sao Paulo in April.

Sales contracts to buy US homes rose last month

(AP) ? The number of Americans who signed contracts to buy homes rose only slightly, suggesting sales may level off in the coming months after solid gains in the past year.

The National Association of Realtors said Thursday that its seasonally adjusted index of sales agreements rose in September to a reading of 99.5. That's up from August's reading of 99.2 but below a two-year high of 101.9 reached in July. Contracts are up 14.5 percent from a year ago.

An index reading above 100 is considered healthy. The index bottomed at 75.88 in June 2010 after a homebuyer's tax credit expired.

The pending home sales index is a measure of the number of signed contracts to purchase homes. The index can signal where the housing market is headed because a signed contract usually results in a final sale one or two months later.

The housing market has been recovering this year, helped by the lowest mortgage rates in decades, a limited supply of homes for sale and steady price increases.

Some economists said the rising trend in home sales will likely continue, despite a few weaker months this fall.

The figures "suggest that existing home sales are likely to flatten over the next month or two," Joseph LaVorgna, an economist at Deusche Bank, said in an email. "Looking beyond then, we expect sales to resume their uptrend as the underlying housing fundamentals continue to improve."

New home sales jumped last month to the highest annual pace in the past two and a half years.

And builders broke ground on new homes and apartments at the fastest pace in more than four years in September. They also requested the most building permits in four years, a sign that many are confident that home sales gains will continue.

The housing recovery could help boost economic growth and hiring at time when manufacturing has weakened sharply. Still, housing makes up a much smaller part of the economy than it did before the bubble. So rising construction and sales may have less of an impact.

And many first-time homebuyers are struggling to qualify for mortgages, since banks have raised credit standards. Many also demand high down payments that many would-be homebuyers can't afford.

Guns, climate, gays missing in presidential race

WASHINGTON (AP) ? Of the roughly 50,000 words spoken in this month's three presidential debates, none were "climate change," ''global warming" or "greenhouse gas."

Housing was discussed in the first debate, but the word "foreclosure" was mentioned in none. Nor was gay marriage.

The 2012 presidential campaign, not just the debates, has focused heavily from the start on jobs, pushing other once high-profile issues to the sidelines. It dismays activists who have spent decades promoting environmental issues, gay rights, gun control and other topics, sometimes managing to lift them to the top tiers of national attention and debate.

With fewer than half of Americans believing that human activity contributes to global warming, according to Pew Research, President Barack Obama talks far less about climate change than he did four years ago. When he locked up the Democratic nomination in June 2008, he said future generations would recall "this was the moment when the rise of the oceans began to slow and our planet began to heal."

Obama hasn't come close to making such claims in recent months.

Last June, 3,100 U.S. temperature records were broken and much of the nation was in drought, said Daniel Kessler, spokesman for 350 Action Fund, which tries to raise awareness of global warming. And yet the three presidential debates, and the sole vice presidential forum, produced "absolute silence on climate science," Kessler said.

He said he believes voters would respond favorably "if candidates put out a bold vision" on climate change. Instead, the topic generates far less discussion than it did four years ago. Other topics suffering downgrades in presidential campaign attention include:

GUNS

Gun rights sometimes have played big roles in U.S. elections. In 1993, for instance, the recently elected President Bill Clinton signed the Brady Bill, which required background checks on many gun buyers. A new ban on assault weapons soon followed. But it contributed to heavy Democratic congressional losses in 1994, and the law expired 10 years later.

Despite high-profile mass shootings ? including those involving Rep. Gabrielle Giffords in early 2011, and dozens of Colorado movie theater patrons in July ? questions about gun rights and gun control have generated scant discussion this year.

They weren't mentioned in the first debate between Obama and Republican Mitt Romney, which focused on domestic issues. A voter asked about assault weapons in the second, town-hall-style debate, on Oct. 16.

Obama cautiously spoke of "seeing if we can get an assault weapons ban reintroduced." Romney said he favors no new gun laws.

The Brady Campaign to Prevent Gun Violence criticized the lack of discussion in the first debate. After the second, it chiefly praised the woman who asked about the assault weapons ban.

IMMIGRATION

In recent years, immigration has been a big issue in national elections. Then-Texas Gov. George W. Bush backed a pathway to legal status for many illegal immigrants when he ran for president in 2000. But Republican activists and members of Congress increasingly took hard stands against illegal immigrants.

In this year's Republican primaries, Romney positioned himself to the right of Texas Gov. Rick Perry, former Speaker Newt Gingrich and others when it came to being tough on illegal immigration. He said illegal immigrants should be encouraged to "self-deport."

Romney has softened his rhetoric somewhat since then.

He told Hispanic leaders in June he would replace Obama's executive order allowing 800,000 young illegal immigrants to remain in the country with a long-term solution. But Romney didn't detail his plans, and immigration has played a smaller role in the general election than it did in the primary.

GAY RIGHTS

The gay rights advocacy group Human Rights Campaign recently noted on its website that questions of same-sex marriage and other gay rights issues were not mentioned in the first two presidential debates and the vice presidential debate. The final Obama-Romney faceoff was likely to be "the last window for the candidates to share their vastly different views" on the topic, the group said.

But that debate, on Oct. 22, also came and went with no mention of "gay" or "same-sex" ? or any type of marriage, for that matter.

That's not to say the nominees haven't taken stands. Early in his term, Obama helped clear the way for gays and lesbians to serve openly in the military. Last May he embraced same-sex marriage, which Romney opposes.

The gay-oriented group Log Cabin Republicans gave Romney a somewhat tepid endorsement. "If LGBT (lesbian, gay, bisexual, transgender) issues are a voter's highest or only priority, then Governor Romney may not be that voter's choice," the Log Cabin Republicans endorsement said. The group said it believed Romney would do a better job of cutting spending and creating jobs.

Those are precisely the issues that have dominated the campaign, often in basic discussions that don't go far beyond platitudes. Both campaigns have warned of the looming "fiscal cliff" ? the package of huge tax hikes and spending cuts scheduled to hit Jan. 1 ? but no one has come close to offering a viable solution.

Meanwhile, advocates for improving schools, fighting poverty, fighting crime and other issues ? which have played big roles in past elections ? have struggled to get a word in edgewise this year.

When U.S. unemployment exceeded 10 percent in October 2009, politicians said the 2012 election would be about jobs, jobs, jobs. Three years later, that hasn't changed.

Individual gene differences can be tested in zebrafish

ScienceDaily (Oct. 25, 2012) ? The zebrafish is a potential tool for testing one class of unique individual genetic differences found in humans, and may yield information helpful for the emerging field of personalized medicine, according to a team led by Penn State College of Medicine scientists. The differences, or mutations, in question create minor changes in amino acids -- the building blocks of DNA -- from person to person. Zebrafish can be used as a model to understand what biological effects result from these genetic mutations.

Personalized medicine uses modern technology and tools to find biological and genetic differences in individuals so that treatment is more effectively delivered.

"A major challenge of personalized medicine is the lack of a standard way to define the importance of each of the many unique mutations found in an individual's genetic code," said Keith Cheng, M.D., Ph.D., professor of pathology and lead researcher. "Approaches are particularly needed to experimentally determine what differences these mutations make. It is difficult to distinguish the effects of a single amino acid change caused by those changes in our DNA."

The zebrafish is a good choice because of its similarity to humans as a vertebrate, its transparency as an embryo and the powerful genetic tools available in this model organism.

The Cheng lab's approach is like testing small damages in car parts, one at a time. For example, a "mutant" car headlight is known not to work when a certain connector is missing. Taking a normally functioning connector out of a working headlight and replacing it with a connector damaged in a specific way -- a cracked wire casing or a corroded wire connector, for example -- can show whether the damage matters. If the light works, then that mutation makes no difference on the function of the headlight. If the light does not work, the mutation has an effect.

Postdoctoral fellow Zurab Tsetskhladze, who performed the zebrafish experiments, tested this method with two genes that affect skin color. He started with an equivalent of the broken car part: mutant zebrafish with lighter pigment cells. First, Tsetskhladze confirmed that by injecting normal messenger RNA (ribonucleic acid) into the mutant zebrafish, the lighter pigment cells become "cured" -- or darker --like those of a normal zebrafish. Messenger RNA makes the cells produce the protein the scientists want to study.

Tsetskhladze was then able to test RNA with only one "human" mutation to see if cure was still possible. Cure suggests that the mutation does not matter. If cure is prevented by the mutation, the conclusion is that the protein's function is affected by the amino acid difference being tested.

Cheng's lab works with zebrafish to study genetic differences that contribute to human skin color. Scientists want to determine the role these differences play in the development of skin cancer, and to find ways to better protect people from cancer.

In the current study, two of the amino acid differences that Cheng has shown in prior studies to contribute to light skin color in humans prevented the zebrafish color from darkening. A third amino acid difference that is common in Eastern Asians was of unknown effect. The researchers found that the change made no difference in function in zebrafish. This finding matched the findings of K.C. Ang, postdoctoral fellow, who found no effect of the tested change on the skin color of East Asians.

To see if this approach might be used in other ways, Stephen Wentzel, graduate student, Penn State College of Medicine, looked at mutations in the four genes known to contribute to albinism, which lightens the color of skin, eyes and hair, and is associated with any one of more than 250 known single amino acid differences. He found that at least 210 of these are theoretically testable in the zebrafish. This new test may help scientists to determine which mutations can be ignored and which may need action -- such as a change in life habit.

"This approach may potentially be extended to other biological functions and may therefore be useful in personalized medicine," Cheng said.

The researchers published their findings in PLoS ONE.
